DELAYED-TYPE HYPERSENSITIVITY (DTH)
in Lewis rat
INTRODUCTION
DTH is immune reaction in which memory T-cells play a pivotal role. The reaction is antigen specific and causes erythema, oedema and induration at the site of antigen injection in immunized animals or humans.

DTH is induced in female Lewis rats by subcutaneous injection of emulsion of ovalbumin/complete Freund's adjuvant. One week later, rats are challenged with injection of ovalbumin in the pinna ear. The DTH reaction is determined by measuring the increase in the pinna ear thickness 24 hours after the ovalbumin challenge.
COMPOUND TESTING
Compound testing addresses the effect of treatment of rats on pinna ear swelling.
Ear swelling induced by ovalbumin challenge is reduced by dexamethasone treatment.
